I don't know why you would use either of those options, both older motors, What drives do you have or plan to use? the 480 Yanmar is a good engine but a better choice would FPT 570 (which can come with 600hp) or the Cummins 6.7 550hp, the FPT has much better packaging though. I have Yanmar 315's (6LP) with Bravo drives in my boat and it is a great setup.
My real problem is I only have 60" from transom to bulkhead, which is why the duramax looks good to me, I don't think I can get the inline six cummins or yanmars in there.

My boat was built for 315's/xr's but I'm thinking that at 10,000#'s and 40' I'm to heavy for the 315's. ( 6200#'s w/o engines,trans,drives,rigging)

If you have 60" from transom to bulkhead you can fit the Yanmar 315's with a foot to spare or you can jam in the Cummins or FPT. The FPT is 54.5" overall but from flywheel to pulleys it is only 43" there is a lot of overhang on top with the turbo. I have the install specs for all three.
My boat is 37' weighs 9000-10000 lbs dry, loaded up with fuel and 10 people it will still run 52-54, just two people and light fuel it will do 57. I have a full hardtop with 4 ft radar, forward seating and forward extra fuel tank. PM me your email and I will send you some photos of my installation. Your boat sounds lighter than mine you might be able to break 60 with the Yanamr 315's. Mine will do it I just have to turn a few screws on the pumps
The Cummins or FPT would be a nice setup but you will have a pretty tail heavy boat and with drivetrain the power package will be triple the cost of a Yanmar 315/Bravo package, and you would be lucky to get 20 mph more.

I still have several hours of fairing and painting to go so I have some time to decide.
I have #6 gimbels and drives for it and wanted 450-500 hp diesels, but they may kill my range and make it stern heavy.
I'm going to sandbag the engine bay and float the hull to check the waterline and attitude next week.
Lot of weight increase with a 5.9 cummins/trans/#6 drive compared to 315/bravoxr, not sure how much power is used up pushing the extra weight. Although I've ridden on Innovation's old 42' fountain test boat that ran 55 WOT with a single duramax and a deep set #6. I had a deposit on the last pair of mechanical 315's Mastry had last summer but I backed out because I was worried I need more HP with this hull, it originally was a mid eighties Hawk 40 and has been completely gutted out and and relaminated with Kevlar/ carbon fiber and a center console stringer grid.

17-18 gph, your not far from Miami come over and I'll take you for a ride, you are about to drop a lot of $$ on a power package might as well see in person. The Cummins or FPT are about 1000 lbs more than my Yanmars but that was using xPower drives, for #6'S probably 1500 more.
