Can I run the standard gear from a gen V non roller 502 on a 3 piece billet cam with an iron gear?

Yes. I ran my original TB IV gear on a Crane Roller cam with a pressed on steel cam gear.
Eddie Young has suggested in another thread that you always start with a new distributor gear when using a new cam. I never have, but its cheap insurance.

I am pretty sure the TB IV is a cast gear at least mine where.
I had some cams made for my new engines, it would have take 2-3 weeks longer to have a cast gear pressed on, some kind of shortage of them. So I went to steel gears.
