You will be able to check the lifter and cam lobe as soon as you pull the intake manifold.

If the intake valve is not opening, then no air is getting in to be compressed. If a valve is stuck open, then air will just escape instead of compressing.

With both valves closed shouldn't I still pull air in and push it out through the spark plug if there is nothing blocking that port?

I saw on MWBP that you found a bent valve. Now you need to find what caused it.
