Default 1 piece crank in a 2 piece block

Looking for a block to replace my cracked gen 6. Found a great deal on a Merlin III. However, it is a 2 piece RMS block. Can a 1 piece crank work in a 2 piece RMS block? I know there are adapters for the reverse set up.

I have a 454 4 bolt main Gen IV bare block, not rusted, std bore..$650...
